Cocaine Activates Genes Through Chromatin Remodeling

ثبت نشده
چکیده

Chromatin remodeling contributes to healthy adaptations, including learning and memory, and to diseases, including schizophrenia. Dr. Eric Nestler and colleagues at the University of Texas Southwestern Medical Center and Harvard Medical School revealed its role in cocaine pathology. In a series of experiments with rats, they demonstrated that cocaine-induced chromatin remodeling increases the production rates of four proteins that have been implicated in cocaine addiction: cFos, ∆ FosB, BDNF, and Cdk5.

Chromatin Remodeling Is a Key Mechanism Underlying Cocaine-Induced Plasticity in Striatum

Given that cocaine induces neuroadaptations through regulation of gene expression, we investigated whether chromatin remodeling at specific gene promoters may be a key mechanism. We show that cocaine induces specific histone modifications at different gene promoters in striatum, a major neural substrate for cocaine's behavioral effects. At the cFos promoter, H4 hyperacetylation is seen within 3...

Parsing molecular and behavioral effects of cocaine in mitogen- and stress-activated protein kinase-1-deficient mice.

Although the induction of persistent behavioral alterations by drugs of abuse requires the regulation of gene transcription, the precise intracellular signaling pathways that are involved remain mainly unknown. Extracellular signal-regulated kinase (ERK) is critical for the expression of immediate-early genes in the striatum in response to cocaine and Delta9-tetrahydrocannabinol and for the rew...

HOXA10 controls osteoblastogenesis by directly activating bone regulatory and phenotypic genes.

HOXA10 is necessary for embryonic patterning of skeletal elements, but its function in bone formation beyond this early developmental stage is unknown. Here we show that HOXA10 contributes to osteogenic lineage determination through activation of Runx2 and directly regulates osteoblastic phenotypic genes. In response to bone morphogenic protein BMP2, Hoxa10 is rapidly induced and functions to a...

Osa-containing Brahma chromatin remodeling complexes are required for the repression of wingless target genes.

The Wingless signaling pathway directs many developmental processes in Drosophila by regulating the expression of specific downstream target genes. We report here that the product of the trithorax group gene osa is required to repress such genes in the absence of the Wingless signal. The Wingless-regulated genes nubbin, Distal-less, and decapentaplegic and a minimal enhancer from the Ultrabitho...

Differential regulation of MeCP2 and PP1 in passive or voluntary administration of cocaine or food.

Cocaine exposure induces changes in the expression of numerous genes, in part through epigenetic modifications. We have initially shown that cocaine increases the expression of the chromatin remodeling protein methyl-CpG binding protein 2 (MeCP2) and characterized the protein phosphatase-1Cβ (PP1Cβ) gene, as repressed by passive i.p. cocaine injections through a Mecp2-mediated mechanism involvi...
